上海交通大学医学院附属仁济医院临床干细胞研究中心,成立于2010年,由仁济医院和上海交通大学Med-X研究院共同创建。研究中心坐落于仁济医院东院5号楼,实验空间3000平方米。截止2018年底,设有研究组5个,科技人员28人。具有高级专业技术职称者13人,研究生导师13人(博导5人、硕导13人),招生专业为肿瘤学,基础医学,现有在读研究生38人(博士生21人、硕士生17 人)。

The Clinical Stem Cell Research Center was established in 2010 and was co-founded by Ren Ji Hospital and Med-X Research Institute of Shanghai Jiao Tong University. The research center is located in Building 5 of the East Campus of Ren Ji Hospital, with an experimental space of 3,000 square meters. By the end of 2018, there were 5 research groups and 28 staff (including 13 associate/full professors, 13 graduate student mentors (5 doctoral supervisors and 13 master supervisor), and 38 graduate students (21 doctoral students and 17 master students).Enrollment majorsoncology, basic medicine science.

研究中心的主要研究方向为干细胞与组织修复;肿瘤干细胞与肿瘤发生发展。五个研究组包括现任中心主任高维强教授领衔的“干细胞与组织修复及肿瘤发生研究室”、冯海忠研究员领衔的“肿瘤转移与肿瘤干细胞研究室”、朱鹤研究员领衔的“成体干细胞和肿瘤干细胞研究室”、薛婧研究员领衔的“炎症与炎症相关肿瘤研究室”和蔡志伟研究员领衔的“前列腺肿瘤生物学与模式创建研究室”。研究涉及干细胞分裂模式和干细胞干性维持、肿瘤干细胞和上皮间质转化、免疫与肿瘤等多个领域。中心拥有激光共聚焦显微镜、流式仪(Aria)、流式仪(Fortessa)、流式仪(C6)、切片扫描仪、X-rad 辐照仪、超速冷冻离心机、定量PCR仪、、等先进科研设备。

The main research areas of the Ren Ji Clinical Stem Cell Research Center are stem cell and tissue repair; tumor stem cells and tumor development. The five research groups include the "Stem Cell and Tissue Repair and Tumor Research Laboratory" led by Professor Wei-Qiang Gao (current director of the Center), the "Tumor Metastasis and Cancer Stem Cell Research Laboratory" led by Haizhong Feng, and the "Tissue Stem Cell and Cancer Stem Cell Laboratory" led by Helen He Zhu, "The Laboratory of Inflammatory and Inflammatory-Related Tumors" led by Jing Xue and the "Prostate Cancer Biology and Disease Model Laboratory" led by Chee-Wai Chua. The research covers stem cell division patterns and stem cell maintenance, tumor stem cell and epithelial-mesenchymal transition, tumor immunity, etc. The Ren Ji Clinical Stem Cell Research Center is very well-equipped with laser confocal microscopes, flow cytometers, slide scanner, X-rad irradiator, ultra-speed refrigerated centrifuge, quantitative PCR, etc. 

成立以来,研究中心发展十分迅速。截止2018年底,先后主持56项国家级项目和42项省部级项目,科研经费突破了1亿元。其中,科技部国家重大科学研究计划/国家重点研发计划项目首席2项,国家自然科学基金重点项目3项,面上项目30项,青年项目18项,外国青年项目2项。科研成效显现,产出了一批有影响的重要科研成果,共发表SCI论文120篇,其中IF≥10的13篇,5≤IF<10的47篇,包括Gastroenterology、Nature Communications、Cell Research、PNAS, Clinical Cancer Research等高质量杂志论文,并申请专利10项,授权2项。

Since its establishment, the Ren Ji Clinical Stem Cell Research center has developed very rapidly. By the end of 2018, it had obtained 56 national-level projects and 42 provincial-level projects. The total research funding exceeded 100 million CNY, including 2 National Key R&D Program of China, 3 key projects from National Natural Science Foundation (NSFC), 30 NSFC regular projects, 18 NSFC youth projects, 2 NSFC foreign youth projects. A number of important scientific research achievements with international influence have been produced. A total of 120 SCI papers have been published (13 papers with IF≥10 and 47 papers with 5≤IF<10, including publications in Gastroenterology, Nature Communications, Cell Research, PNAS, Clinical Cancer Research and other high-profile scientific journals). In addition, 10 patents have been filed2 authorized patents

 

    中心注重队伍建设和人才培养。先后从美国、加拿大、德国、英国、日本、马来西亚等国家和地区引入6名科研人才来中心工作,有10人次先后入选了国家和上海市的各类人才计划,其中包括国家特聘专家1人、“王宽诚”讲席教授1人、东方学者6人、青年东方3人、上海青年拔尖2名,上海市卫生系统新优青1人,浦江人才4人、上海市青年科技启明星3名,上海市青年科技英才杨帆计划1名。目前中心已出站10名博士后、毕业20名博士生和14名硕士生,培养了一批(10多人)的博士生,成为赴美国进行博士后研究的青年才俊,培养他们的学术追求志向,分别去美国哈佛(2名)、耶鲁、哥伦比亚、华盛顿大学、西北大学、NIH、贝勒医学院(2名),MD Anderson 癌症研究中心,纪念斯隆-凯特琳癌症中心、UCSD博士后,2名硕士生去了欧洲海德堡大学、柏林自由大学攻读博士。先后有多人次荣获各类国家和上海市基金和奖项,其中包括国家博士后基金2人、上海市博士后基金1名,国家奖学金6人。

 The center makes great effort in team building and talent recruitment. Six principle investigators have been recruited from the United States, Canada, Germany, the United Kingdom, Japan, Malaysia. Ten staff have been selected into various national and Shanghai talent programs, including 1 National Distinguished Expert, 1 KC Wong Professor, 6 Oriental Scholars, 3Youth Oriental Scholars, 2 Shanghai Youth Talent Support Program, 1Excellent youth scholar in shanghai health system4 Pujiang Talents, 3 Shanghai Science and Technology Rising-Stars, 1 Shanghai Science and Technology Yang Fan project. The center has already trained 10 post-doctors, 20 doctoral students and 14 master students. More than 10 PhD students were trained to become young talents with high academic aspirations for thier post-doctoral research in several famous universities in United States including Harvard (2 persons), Yale, Columbia, Washington University, Northwest University, NIH, Baylor Medical College (2 persons), MD Anderson Cancer Research Center, Memorial Sloan-Kettering Cancer Center and UCSD, respectively. Two Master students went to the University of Heidelberg and Freie Universit?t Berlin (FU Berlin) in Europe for their PhD degree, respectively. Many of them successfully applied national and Shanghai research funds or awards, including 2 national post-doctoral funds, 1 Shanghai post-doctoral fund and 6 National scholarships.

中心也重视国内外学术活动和合作交流。先后组织召开和参与了一系列重要国际、国内学术会议。包括举办“干细胞分裂模式和干细胞干性维持国际学术论坛”、“干细胞与癌症研究前沿国际会议”等

The Clinical Stem Cell Research Center values both national and international scientific cooperation and exchanges. It has hosted multiple important academic activities and conferences, including the " international forum on stem cell division mode and stemness maintenance " "Frontiers in stem cell and cancer research international conference" and so on.

 

  未来十年,中心将基于现有研究方向,紧密结合临床,整合与优化现有学科平台,研究干细胞与发育和组织修复、肿瘤干细胞与肿瘤发生发展,寻找新的干细胞和肿瘤生物标志物、分子分型及肿瘤诊断治疗靶标,建成在国内外有影响力的临床干细胞和肿瘤基础研究与临床转化基地。 

In the next ten years, the Ren Ji Clinical Stem Cell Research Center will aim to further integrate its existing academic platforms and the clinical strength from Ren Ji hospital to study stem cells in development and tissue repair, tumor stem cells and tumorigenesis, to uncover new stem cells and tumor biomarkers, to provide new approaches for molecular classification of cancer, and to develop novel cancer diagnostic and treatment targets. The research center will strive to build a base with domestic and international influence for stem cell and cancer basic research and clinical translation.

                    The hallmarks of solid tumor are their highly proliferation, infiltrative, and cancer stem-like cell (CSC) self-renewal. We are studying the signaling cascades underlying the enhanced tumor progression and CSC self-renewal capacity of glioma and breast cancer. Our goal is to understand all aspects of glioma and breast cancer including the microenvironment/niche, molecular markers, and self-renewal pathways, and develop novel therapeutic strategies targeting glioma and breast cancer cells to improve clinical treatment.

大家课题组的主要研究方向,包括探究前列腺上皮和肿瘤细胞雄激素受体(AR)非依赖特质的内在与外在调节机制。在前期的研究中,大家成功鉴定了具备AR非依赖特质的前列腺上皮细胞,并将进一步评估这些上皮细胞亚群的祖细胞特质,以及作为恶性前列腺肿瘤起源细胞的能力。大家希翼这些研究工作最终能为AR非依赖型前列腺癌病患带来全新的治疗策略。同时,课题组将致力优化之前建立的类器官培养系统,以用于培养不同类型的恶性肿瘤,包括AR非依赖型前列腺癌、膀胱腺癌、转移性卵巢癌和三阴性乳腺癌。大家将通过前沿的力学与化学科学手段,鉴定不同肿瘤类型的类器官培养条件,并将不同的基质细胞成分整合到培养系统中,以探讨基质与肿瘤细胞之间的相互作用。

Our group studies the intrinsic and extrinsic regulation of androgen receptor (AR) independence in normal and malignant prostatic tissues. In particular, we have identified and characterized novel AR-independent epithelial stem cell or progenitor populations, and are currently investigating their ability to serve as the cell of origin for prostate cancer. Ultimately, we aim to develop novel therapeutic strategies for the lethal AR-independent prostate cancer. The second goal of our group is to extend the use of an organoid assay that we established previously to grow aggressive tumor types, include AR-independent prostate cancer, bladder adenocarcinoma, metastatic ovarian cancer and triple negative breast cancer. We aim to identify specialized organoid culture conditions for different tumor types and to integrate stromal components into the culture system using cutting-edge mechanical and chemical approaches.
