【报告题目】Non-invasive optical characterization of cells and tissues using endogenous contrast
Disruption of cell-matrix interactions is associated with a wide range of diseases such as cancer, fibrosis, and neurodegenerative diseases. Modified interactions have major effects upon key functional cellular features, as well as upon the matrix biophysical and biochemical properties. In this talk, I will focus on my recent work of improved characterization of these structure-function connections based on endogenous contrast of cells and tissues. With regards to the cellular features, we use two-photon excited fluorescence (TPEF) microscopy to acquire images from conenzymes of NADH and FAD which are actively involved in a number of metabolic pathways, and develop novel optical biomarkers to characterize cell features, including the optical redox ratio, phasor fluorescence lifetime and mitochondrial clustering. On the other hand, we use second harmonic generation (SHG) approach to image collagen fibers in a 3D context. We develop 3D orientation and organization algorithms to characterize the spatial alignment of collagen, which proves to be a sensitive biomarker in several diseases, such as cancer and traumatic brain injury. Overall, these improvements in cell and matrix characterization lead to a better understanding of cell-matrix interactions.
【报告人概况】刘智毅，博士，2013年获清华大学理学博士学位，之后进入美国塔夫茨大学澳门太阳集团娱乐8722工程系开展博士后研究至今，导师为Irene Georgakoudi教授。主要研究方向为澳门太阳集团娱乐8722光子学，包括双光子荧光成像，荧光寿命成像，二次谐波成像等技术及在疾病诊断中的应用，并发展了相应的图像处理方法和表征参量。已发表SCI论文20余篇，其中包括Biomaterials, Biosensors & Bioelectronics等第一编辑或通讯编辑的论文10篇；在合作编辑论文中，包括Science Translational Medicine 的封面文章等。担任Chemical Communications, Journal of Biophotonics, Biomedical Optics Express等杂志的审稿人。
